This dataset supports the Master of Philosophy thesis entitled "Ethnopharmacological Strategy for Autoimmune Regulation: Cellular Mechanisms and Key Molecular Targets in Sjögren's Syndrome," submitted by Xiaoyu Xu at The University of Hong Kong in August 2025. The research systematically investigates the immunomodulatory effects and therapeutic potential of the classical traditional Chinese medicine formula Yu Ping Feng (YPF) and its key bioactive constituent Atractylenolide I (AT1) in the context of primary Sjögren's syndrome (SS), an autoimmune disorder characterized by exocrine gland dysfunction and systemic immune dysregulation.The dataset includes experimental results from:* Network pharmacology analyses identifying molecular targets and pathways of YPF and AT1 related to immune regulation in SS;* In vivo validation using an experimental SS mouse model assessing salivary gland function, histopathology, autoantibody levels, and immune cell subset composition;* In vitro assays examining the modulatory effects of AT1 on T follicular helper (Tfh) cell differentiation and function in both mouse and human CD4+ T cells;* Transcriptomic profiling and functional validation of the KLF2 signaling pathway as a key molecular mechanism mediating AT1’s immunoregulatory action;* Clinical pilot data from SS patients treated with YPF demonstrating improved clinical symptoms and immune biomarkers.The data comprehensively covers multi-dimensional immune phenotyping, molecular pathway analysis, and therapeutic evaluation, providing a valuable resource for understanding the mechanisms of traditional Chinese medicine in autoimmune disease modulation. Sensitive or identifiable patient information is excluded to protect privacy, and all experimental procedures were conducted following approved ethical guidelines.
