We observed three different matrix-producing variant colonies of Escherichia coli UTI89 population from the biofilm pellicle, when plated on congo red (CR) agar. One among which was a heterogeneous colony variant (HCV), which displayed red dry and rough (RDAR) morphology in the middle layer of the colony, while the peripheral cells displayed white color on CR agar plate. This implied that centre of the colony produces cellulose and curli and the peripheral cells did not. The other two variants that were found were called as red colony variant (RCV) and white colony variant (WCV). In the study, we performed competition studies between the variants and found that the interactions are context dependent.
