MiR-9 plays an irreplaceable role in regulating the progression of breast cancer. In order to study the role of miR-9 in promoting MDSC recruitment in the tumor microenvironment, we need to explore the genetic process of miR-9 regulation. We used microarrays to detail the global program of miR-9 down-regulated gene expression and identified different categories of regulated genes in this process. Overall design: A breast cancer cell line with stable knockdown of miR-9 was constructed by lentivirus infection for RNA extraction and Affymetrix microarray hybridization. The experimental group and the control group were repeated 3 times.

微小RNA-9(miR-9)在调控乳腺癌进展过程中发挥着不可替代的作用。为探究miR-9在肿瘤微环境中促进髓系来源抑制细胞(myeloid-derived suppressor cells, MDSC)招募的功能,本研究需阐明miR-9调控的遗传过程。本研究通过基因芯片(microarray)技术详细解析了miR-9下调基因的全局表达谱,并鉴定了该调控过程中不同类别的受调控基因。整体实验设计:通过慢病毒感染构建稳定敲低miR-9的乳腺癌细胞系,用于RNA提取及Affymetrix基因芯片杂交。实验组与对照组均设置3次生物学重复。
