Objective: To investigate the effect of hypoxia on the migration of mesenchymal stem cells (MSCs) and the role and mechanism of the Yiqi Huoxue formula in regulating the migration of hypoxic MSCs.Methods: After performing a scratch wound assay, cells were subjected to hypoxia in a hypoxic chamber. The experimental groups were as follows: N0 group (normal culture for 24h without migration), N1 group (migration state under normal culture for 24h), M1 group (hypoxic migration for 24h), M2 group (hypoxic migration for 6h), YH1 group (treated with Yiqi Huoxue formula for 24h).Total RNA was extracted using Trizol, and transcriptome sequencing was performed.
研究目的:探讨缺氧对间充质干细胞(mesenchymal stem cells, MSCs)迁移能力的影响,以及益气活血方(Yiqi Huoxue Formula)调控缺氧MSCs迁移的作用与机制。
实验方法:首先开展划痕愈合实验,随后将细胞置于缺氧培养箱中进行缺氧处理。实验分组如下:N0组(正常培养24h,未进行迁移实验)、N1组(正常培养条件下处于迁移状态24h)、M1组(缺氧处理并迁移24h)、M2组(缺氧处理并迁移6h)、YH1组(经益气活血方处理24h)。采用Trizol试剂提取总RNA,并进行转录组测序。
