The aim of this study was to investigate the effect of double knockout versus double heterozygous of Apoe and Clu in mice. Hemi-brains from mice (ACluE14 and ACluE18) with indicated genotypes were perfused and harvested. The brains samples were subjected to 10x Genomics single cell RNA sequencing analysis. The libraries were sent to Genome Analysis Core at Mayo Clinic for sequencing using illumina HiSeq4000 platform.

本研究旨在探究小鼠中载脂蛋白E(Apoe)与簇集素(Clu)双基因敲除与双杂合子两种基因型的效应差异。取携带指定基因型的ACluE14与ACluE18小鼠的半脑,经灌注处理后取材。将所得脑组织样本开展10x Genomics单细胞RNA测序分析。构建完成的测序文库被送至梅奥诊所(Mayo Clinic)基因组分析核心(Genome Analysis Core),采用Illumina HiSeq4000测序平台完成测序。
