Adenylate kinase 4 (AK4) is localized in the mitochondrial matrix, and is believed to be involved in stress, drug resistance, the malignant transformation of cancer, and ATP regulation. We produced AK4 knockdown HeLa cells by using shRNA and used these to analyze resistance, and found that sensitivity to hypoxia and drugs increased. Overall design: 3 samples of HeLa cells haboring AK4 shRNA plasmids and 4 samples of HeLa cells haboring control shRNA plasmid.

腺苷酸激酶4(Adenylate kinase 4,AK4)定位于线粒体基质,被认为参与应激反应、耐药性、癌症恶性转化及ATP调控。本研究通过短发卡RNA(short hairpin RNA,shRNA)构建了AK4敲低的海拉细胞(HeLa),并利用该细胞模型开展耐药性分析,结果发现细胞对缺氧及药物的敏感性均显著升高。实验设计概况:携带AK4 shRNA质粒的海拉细胞样本共3份,携带对照shRNA质粒的海拉细胞样本共4份。
