Gene expression signatures in GIST-T1 cells transfected with a miR-335 mimic molecule

To clarify the effect of miRNAs, we carried out a gene expression microarray analysis using GIST-T1 cells transfected with a miR-335 mimic or a negative control. We found that 1,095 probe sets (853 unique genes) were downregulated (>1.5-fold) by ectopic miR-335 expression. GIST-T1 cells were transfected with a mirVana miR-335 mimic (Ambion) or a mirVana miRNA mimic Negative Control #1 (Ambion). Forty-eight hours after transfection, total RNA extraction was carried out, and gene expression signatures were analyzed.
为阐明微小核糖核酸(miRNAs)的调控效应,我们采用转染了miR-335模拟物或阴性对照的GIST-T1细胞开展基因表达微阵列分析。结果显示,异位表达miR-335可使1095个探针集(对应853个独特基因)的表达水平下调超过1.5倍。本实验中,GIST-T1细胞分别转染了mirVana miR-335模拟物(Ambion)及mirVana miRNA模拟物阴性对照#1(Ambion);转染48小时后,我们提取总RNA并分析基因表达特征。
