This study was carried out to investigate the gene expression profile from arm and hatchlings transcriptome across multiple individual Octopus bimaculoides . 3 different arm sections from an adult male Octopus bimaculoides were collected (containing all tissues, including skin, sucker, muscle and nerve). 2 arm tips were collected from a different adult male and one adult female. One whole-body 30 day old hatchling was removed from chorion and anesthetized. All animals were anesthetized in ethanol, all samples were dissected in sea water and flash frozen. RNA was extracted using Trizol and DNAse treated to carry out Ribo-Zero library preparation.
本研究旨在探究多只双斑蛸(Octopus bimaculoides)的腕足与幼体转录组的基因表达谱。研究收集了1只成年雄性双斑蛸的3个不同腕足区段,该样本涵盖皮肤、吸盘、肌肉及神经等所有组织;另从1只不同的成年雄性与1只成年雌性个体各采集2个腕尖样本。此外,选取1只30日龄的完整幼体,将其从卵壳中剥离并麻醉。所有实验动物均经乙醇麻醉,所有样本均在海水中解剖并快速冷冻。采用Trizol法提取RNA,经DNA酶处理后进行Ribo-Zero文库制备。
