To gain insight into patterns of chromatin accessibility in distinct cell subtypes of mouse pituitary, pituitaries from two adult mice were collected and subjected to single-cell ATAC-seq analysis using 10X Genomics platform Two adult pituitaries were dissected and processed indiviudally for single-cell ATAC-seq. Nuclei were isolated and diluted to achieve ~6000 individual nuclei for library construction. Libraries were sequenced to ~50% saturation. Analysis was done using Cell Ranger pipeline.
为解析小鼠垂体不同细胞亚型的染色质可及性模式,本研究收集了两只成年小鼠的垂体组织,采用10X Genomics平台开展单细胞ATAC-seq(single-cell Assay for Transposase-Accessible Chromatin using sequencing)分析。实验具体步骤如下:对两只成年小鼠的垂体分别进行解剖分离并独立处理;分离细胞核并稀释至约6000个单个细胞核用于文库构建;将文库测序至约50%的饱和率;数据分析采用Cell Ranger流程完成。
