The 3-leaves seeding were watered using the solution with γ-PGA (10 KD, 50 mg/L) and NaCl (100mM). And the control group was only watered with NaCl (100mM) solution. After growth for 20 days, the soils tightly bound to the roots (served as rhizosphere soils) were collected and analyzed for the composition of microbial community. Total bacterial genomic DNA samples were extracted using OMEGA Soil DNA Kit (D5625-01, USA) This experiment was performed in triplicate. 本实验采用γ-聚谷氨酸(γ-PGA,分子量10 KD,浓度50 mg/L)与氯化钠(NaCl,100 mM)的混合溶液浇灌三叶期幼苗,对照组仅用100 mM氯化钠溶液浇灌。幼苗生长20天后,收集紧密附着于根系的土壤(即根际土壤),并分析其微生物群落组成。采用OMEGA土壤DNA提取试剂盒(D5625-01,美国)提取所有细菌基因组DNA样本。本实验设置三次生物学重复。随后对玉米根际土壤细菌的16S rDNA进行扩增并开展高通量测序。
