This project is “Medical proteome analysis of osteoporosis- and bone mass-related proteins using the Kibo Japanese Experiment Module of International Space Station (ISS)”. Applying proteomic technologies, this research aims to identify proteins involved in space flight-induced osteopenia as well as osteoporosis occurring on the ground by searching serum, bone and skeletal muscle (soleus, extensor digitorum longus) proteomes. The goal of this study is to get better understanding to protect the health of both astronauts and osteoporosis patients.
Proteome analyses of skeletal muscles from 10 male C57BL/6J mouse (1-g gravity and 3-g gravity [n=5, respectively]) were carried out tryptic digestion followed by LC-MS/MS analysis (DDA). The obtained results were searched against UniProt DB by Mascot, and the identified peptides were filtered at 1% FDR threshold by searching against the randomized decoy DB. 本研究采用蛋白质组学(proteomics)技术,通过分析血清、骨骼及骨骼肌(比目鱼肌(soleus)、趾长伸肌(extensor digitorum longus))的蛋白质组,旨在鉴定航天飞行诱导的骨质减少症,以及地面人群中发生的骨质疏松症相关蛋白。
本研究的核心目标是深化对宇航员与骨质疏松患者健康保护相关机制的认知。
本研究对10只雄性C57BL/6J小鼠的骨骼肌样本开展蛋白质组学分析,实验分为1g重力组与3g重力组,每组各5只(n=5)。
实验流程为先进行胰蛋白酶酶解,随后采用液相色谱-串联质谱(Liquid Chromatography-Tandem Mass Spectrometry, LC-MS/MS)的数据依赖型采集(Data-Dependent Acquisition, DDA)模式完成检测。
所得结果通过Mascot软件比对UniProt(Universal Protein Resource)数据库,并通过比对随机诱饵数据库,以1%的错误发现率(False Discovery Rate, FDR)阈值对鉴定得到的肽段进行过滤筛选。
