Genome sequencing of Psychotria homalosperma was performed by using Roche 454 reads. Specifically, total genomic DNA of Psychotria homalosperma was extracted from fresh leaf collected from Sekimon (26?40'11.3"N 142?09'16.4"E) on Hahajima Island of the Bonin Islands, using the DNasy Plant Mini Kit. The extracted DNA of P. homalosperma was pyrosequenced using a 454 GS Junior System. Multiplex Identifier (MID) tags were used for multiplexing of this sample and another species in the Bonin Islands, then both samples were mixed. Row data were demultiplexed and MID tags removed from the reads using Newbler.
