In this study, we have investigated the relationship between genome-wide DNA methylation pattern and HIV infection using the methylated DNA immunoprecipitation - microarray method. A pair of monozygotic twins was recruited: one of the twins was infected with HIV while the other was not. The immunoprecipitated CpG-methylated DNA from HIV positive subject (test) and from HIV negative subject (input control) were labelled with Cy5 and Cy3 respectively The methylated genomic DNA fragments from HIV+ and HIV- twins were hybridized on the same chip since the identical genetic background

本研究采用甲基化DNA免疫沉淀-芯片(methylated DNA immunoprecipitation - microarray)技术,探究了全基因组DNA甲基化模式与人类免疫缺陷病毒(HIV, Human Immunodeficiency Virus)感染之间的关联。本研究招募一对同卵双胞胎受试者,其中一名个体感染HIV,另一名未受感染。研究人员分别以Cy5与Cy3荧光染料,对HIV阳性受试者(测试组)及HIV阴性受试者(输入对照组)的免疫沉淀CpG甲基化DNA进行标记。鉴于二者具备完全一致的遗传背景,因此将HIV阳性与阴性双胞胎的甲基化基因组DNA片段置于同一张芯片上完成杂交。
