The telsons of four specimens of the Mexican scorpion Paravaejovis schwenkmeyeri were dissected, from which total RNA was extracted. A cDNA library was constructed that was further subjected to RNA-seq by Illumina. The reads were assembled de novo with Trinity and annotated with Trinotate. A total of 8,858,816 reads were obtained, which assembled into 77,108 putative transcripts. From them, 44,043 where identified matching gene sequences in databases. A subgroup was annotated, of which 6,979 matched known arthropod genes. A total of 549 putative transcripts were identified with sequence similarity to annotated genes from arachnids, 289 of which corresponded specifically to scorpions.
本研究对4份墨西哥蝎子Paravaejovis schwenkmeyeri标本的尾节进行了解剖,从中提取总RNA。构建cDNA文库后,采用Illumina平台开展RNA测序(RNA-seq)。利用Trinity软件对测序reads进行从头组装,并通过Trinotate软件完成功能注释。本次测序共获得8,858,816条reads,最终组装得到77,108条推定转录本。其中44,043条转录本可与数据库中的基因序列匹配。对其中一个亚组进行注释后,发现6,979条转录本与已知节肢动物基因匹配。最终共鉴定出549条与蛛形纲动物已注释基因序列相似的推定转录本,其中289条特异性对应于蝎子类基因。
