Histone modification maps of H3K4me3 and H3K27me3 of meiocytes were obtained by microlibrary and high-throughput sequencing by extracting meiocytes from rice varieties Azucena and IR64. And the DNA methylation results of meiocytes and flag-leaf of Azucena and IR64 were obtained. By collating this data, the male meiocyte-specific epigenomic landscape is constructed Overall design: By micromanipulation technology, the meiocytes of Azucena and IR64 were obtained, respectively. Libraries were constructed by CUT&Tag technology to obtain histone modification data. DNA methylation libraries were constructed by scBS-seq technology. In addition, flag-leaf's DNA methylated library is constructed by directly extracting whole genome DNA
本研究从水稻品种Azucena和IR64中分离减数分裂细胞,通过微文库构建与高通量测序,获取了这些减数分裂细胞的H3K4me3与H3K27me3组蛋白修饰图谱;同时还获取了Azucena与IR64的减数分裂细胞及剑叶的DNA甲基化数据。通过整合上述数据,构建了雄性减数分裂细胞特异性表观基因组图谱。
总体实验设计:采用显微操作技术分别分离得到Azucena与IR64的减数分裂细胞;利用CUT&Tag技术构建文库以获取组蛋白修饰数据;采用单细胞亚硫酸氢盐测序(single-cell bisulfite sequencing, scBS-seq)技术构建DNA甲基化文库;此外,通过直接提取全基因组DNA的方式,构建了剑叶的DNA甲基化文库。
