In 3D cultures of hPSC-derived lung progenitors withdrawal of the GSK3 inhibitor, CHIR99021, induces multilineage maturation, including of proximal cells, type I alveolar epithelial cells and morphologically mature type II cells. Lung Progenitors and D50 differentiated cells mRNA profiles were generated by deep sequencing, in triplicate, using Illumina HiSeq2500. D50 differentiated lung progenitor single-cell RNA profiles were generated using HiSeq4000 and 10x genomics' cellranger pipeline v2.0.0.
在人类多能干细胞(human pluripotent stem cell, hPSC)来源的肺祖细胞三维培养体系中,撤除糖原合成激酶3(glycogen synthase kinase 3, GSK3)抑制剂CHIR99021,可诱导多谱系成熟,涵盖肺近端细胞、I型肺泡上皮细胞以及形态学成熟的II型肺泡上皮细胞。肺祖细胞与分化第50天的细胞的mRNA转录组谱,通过Illumina HiSeq2500测序平台完成了三次生物学重复的深度测序构建。分化第50天的肺祖细胞单细胞RNA转录组谱,则依托HiSeq4000测序平台及10x Genomics的Cell Ranger分析流程v2.0.0完成构建。
