Comprehensive downstream mapping of HNF4α revealed its role in regulating GC metabolism via regulation of IDH1 Overall design: HNF4α ChIP-seq of GC cell lines, RNA-seq data of HNF4α perturbation and transcritome primary GC dataset were integrated to reveal HNF4α regulating biological processes and targets in GC, thus provide new therapeutic opportunity.
肝细胞核因子4α(Hepatocyte Nuclear Factor 4α, HNF4α)的全面下游调控图谱揭示了其通过调控异柠檬酸脱氢酶1(Isocitrate dehydrogenase 1, IDH1)参与胃癌(Gastric Cancer, GC)代谢调控的功能。整体实验设计:整合胃癌细胞系的HNF4α染色质免疫沉淀测序(Chromatin Immunoprecipitation Sequencing, ChIP-seq)数据、HNF4α基因扰动后的RNA测序(RNA Sequencing, RNA-seq)转录组数据以及原发性胃癌转录组数据集,以解析HNF4α在胃癌中调控的生物学过程与靶标基因,进而为胃癌治疗提供全新的潜在治疗契机。
