TMV-resistant (N), -susceptible (n), and enhanced susceptible mutant sun1-1 (N) tomato were grown in asceptic conditions in growth chambers (25º, 16hr/8hr light dark). 4-5 week old tomato were treated with TMV leaf sap or mock leaf sap, and leaf tissue was collected after 3, 9, and 27 hours. RNA was isolated from each sample (leaf tissue from one plant/one treatment/one timepoint), using standard TIGR protocols, and 25ug of total RNA was labeled using TIGR indirect labeling protocols. 抗烟草花叶病毒(TMV)型(N)、易感型(n)以及增强易感型突变体sun1-1(N)番茄,均在无菌条件下于生长箱中培养,培养参数为25℃、16小时光照/8小时黑暗。将生长至4~5周龄的番茄分别用TMV叶片汁液或空白叶片汁液进行处理,并分别于处理后3小时、9小时、27小时采集叶片组织。采用标准TIGR实验方案,从每份样本(单株植物、单处理条件、单时间点对应的叶片组织)中提取RNA,并使用TIGR间接标记方案对25μg总RNA进行标记。本实验共重复三次。关键词:直接比较 整体设计:总计30次杂交反应。
