We identified dactylellinatone, a novel nematode chemoattractant from <i>D. haptotyla</i> that enhances fungal predatory efficiency. Heterologous expression and isotope-labeled feeding experiments confirmed that the <i>Dlt</i> gene cluster is directly responsible for the biosynthesis of dactylellinatone. We discovered that the expression of <i>Dlt</i> gene cluster was significantly upregulated during predation. Deletion of the core gene in the cluster not only disrupted dactylellinatone production but also reduced <i>D. haptotyla</i>'s attraction to nematodes and its pathogenicity. Collectively, our work reveals that <i>D. haptotyla</i> can increase their predation efficiency when sensing the presence of nematodes by triggering the expression of <i>Dlt </i>gene cluster and initiating the synthesis of dactylellinatone.
