We used RNA-seq to determine transcriptional profiles of whole guts or IPCs isolated from guts infected with wild type or type VI secretion system deficient Vibrio cholerae. We found significant differences between guts and progenitor cells infected wild type or type VI secretion system deficient Vibrio cholerae. Newly eclosed female flies were raised for 5 days at 29C and then mock infected (with sterile bacterial growth media) or infected with wild Vibrio cholerae (C6706) or type VI secretion system deficient Vibrio cholerae for 24 hours. After 24 hours guts were dissected. The transcriptome of whole guts or intestinal progenitor cells only was sequenced.
本研究采用RNA测序(RNA-seq)技术,对感染野生型霍乱弧菌(Vibrio cholerae)或VI型分泌系统缺陷型霍乱弧菌的完整肠道,以及从该感染肠道中分离得到的肠道祖细胞(intestinal progenitor cells, IPCs)的转录组谱进行测定。研究发现,感染上述两种菌株的肠道与肠道祖细胞之间存在显著的转录组差异。将新羽化的雌性果蝇置于29℃环境中饲养5天,随后分为三组:分别采用无菌细菌培养基进行空白模拟感染,或感染野生型霍乱弧菌(C6706菌株)、VI型分泌系统缺陷型霍乱弧菌,感染时长均为24小时。感染24小时后,解剖获取果蝇肠道,随后对完整肠道或仅肠道祖细胞的转录组进行测序。
