Nicotinamide and pyridoxine have been identified as potent modulators of Muscle Stem cell activity. We characterized genome-wide expression profiles of NAM/PN-treated MuSC and Vehicle to verify if NAM/PN supplementation reverses MuSC dysfunction during Aging. Overall design: Freshly isolated MuSC from aged mice were seeded and treated with either 1% DMSO (vehicle) or the combination of nicotinamide and pyridoxine for 72 hours in growth medium. From 8 mice, we performed 4 independent biological replicates (pool of 2 mice).

烟酰胺(Nicotinamide)与吡哆醇(pyridoxine)已被鉴定为肌肉干细胞(Muscle Stem Cell, MuSC)活性的强效调节剂。本研究对经NAM/PN处理的肌肉干细胞及载体对照组(Vehicle)开展了全基因组表达谱分析,旨在验证NAM/PN补充是否可逆转衰老过程中的肌肉干细胞功能障碍。实验整体设计:从老年小鼠中新鲜分离得到肌肉干细胞,接种后分别采用1%二甲基亚砜(DMSO)作为载体对照,或烟酰胺与吡哆醇的联合处理液,在生长培养基中培养72小时。本实验使用8只小鼠,设置4次独立生物学重复(每2只小鼠的细胞混合为一个重复样本)。
