We used scRNA-seq to understand how CARD9 R101C remodels skin immunity and cell state in response to fungal infection in a mouse model of dermatophytosis. Overall design: We performed single cell RNA sequencing (scRNAseq) of skin from WT and CARD9 R101C mice infected with T. rubrum at day two (D2) and day nine (D9) post-infection (3 replicates of each WT and CARD9 R101C at each timepoint)
本研究采用单细胞RNA测序(single-cell RNA sequencing, scRNA-seq)技术,以探究皮肤癣菌病小鼠模型中,CARD9 R101C突变如何重塑皮肤免疫与细胞状态以应对真菌感染。总体实验设计:我们对感染红色毛癣菌(T. rubrum)的野生型(Wild Type, WT)及CARD9 R101C突变型小鼠,分别于感染后第2天(D2)与第9天(D9)采集皮肤样本进行单细胞RNA测序;每个时间点下,WT组与CARD9 R101C组均设置3次生物学重复。
