Molecular classification of tumour cells in a patient with intravascular large B-cell lymphoma

Intravascular large B-cell lymphoma (IVLBCL) is a rare type of extranodal LBCL. It is characterized by the proliferation of tumour cells exclusively intraluminally in small blood vessels of different organs. We used microarray-based analysis of gene expression of tumour cells isolated from a patient with primary manifestation of the lymphoma in the skin and compared it with various other diffuse LBCLs (DLBCLs) as well as a previously published DLBCL classifier. CD20+CD45+HLA-DR+ tumor cells were FACS sorted from a lesional skin biopsy of the patient and compared to FACS sorted tumor cells from (1) affected lymph nodes from patients with DLBCL (3 samples from patients with DLBCL, nonGCB type; three samples from patients with DLBCL, GCB type) and (2) tumor cells from two patients with primary cutaneous LBCL, leg type.

血管内大B细胞淋巴瘤（Intravascular large B-cell lymphoma, IVLBCL）是一类罕见的结外大B细胞淋巴瘤，以肿瘤细胞仅在多器官小血管管腔内增殖为核心特征。本研究对1例以皮肤为淋巴瘤原发受累部位的患者所分离的肿瘤细胞开展基于微阵列的基因表达分析，并将该分析结果与多种其他弥漫性大B细胞淋巴瘤（diffuse LBCLs, DLBCLs）以及已发表的DLBCL分类器进行比对。研究人员从该患者的皮损皮肤活检组织中通过荧光激活细胞分选术（Fluorescence-Activated Cell Sorting, FACS）分选出CD20+CD45+HLA-DR+的肿瘤细胞，并将其与以下两类样本的分选肿瘤细胞进行对照：（1）弥漫性大B细胞淋巴瘤患者的受累淋巴结样本（3例非生发中心B细胞样亚型DLBCL样本、3例生发中心B细胞样亚型DLBCL样本）；（2）2例原发性皮肤腿型大B细胞淋巴瘤患者的肿瘤细胞。
