MicroRNAs are important cellular components and their dysfunctions are associated with various disease. Acute myocardial infarction (AMI) is one of the most serious cardiovascular diseases. Although several miRNAs have been reported to be associated with AMI, more novel miRNAs are needed to be investigated to ascertain if they are associated with AMI. SD rats (180-200g) was divided into sham-control group and two days group after AMI, seven days group after AMI, fourteen days group after AMI, each group has six individual animals total RNA was taken from the border-zone myocardium , low molecular weight RNA was seperate and labeled , and then hybridized to capitalbio V2 biochip representing about 924 microRNA . three chip were test in each group, and the procedure was repeated twice.
微小RNA(microRNAs)是一类重要的细胞组分,其功能异常与多种疾病相关。急性心肌梗死(Acute Myocardial Infarction, AMI)是最严重的心血管疾病之一。尽管已有多项研究报道多种微小RNA与急性心肌梗死存在关联,但仍需对更多新型微小RNA进行探究,以明确其与AMI的相关性。本研究选用体重180~200g的SD大鼠,分为假手术对照组、急性心肌梗死造模后2天组、7天组及14天组,每组各6只实验动物。从各组大鼠的梗死边缘区心肌组织中提取总RNA,分离低分子量RNA并进行标记,随后与覆盖约924种微小RNA的capitalbio V2生物芯片进行杂交。每组设置3张芯片开展检测,且整个实验流程重复两次。
