The goal of the study is the development of a high-throughput sequencing-based mismatch detection assay (Diff-seq). Diff-seq relies on the activity of Surveyor endonuclease on mismatched molecules, generated after cross-annealing of a complex pool of DNA fragments. Sequencing libraries prepared after enrichment for Surveyor-cleaved molecules result in increased coverage at the variant sites.
本研究旨在开发一种基于高通量测序(high-throughput sequencing)的错配检测分析法(Diff-seq)。Diff-seq 依托Surveyor核酸内切酶(Surveyor endonuclease)对错配分子的酶活性,此类错配分子由复杂DNA片段库经交叉退火后生成。对经Surveyor核酸内切酶切割的分子进行富集后制备的测序文库,可在变异位点(variant sites)获得更高的测序覆盖度。
