MicroRNA has the potential for cross-regulation and functional integration of discrete biological processes during complex physiological events. In this study, we found that the highly expressed microRNAs in exosomes from bone marrow derived macrophage appear to control fibrotic healing response in the tendon. Notably, fibrotic microRNA-21 in mice distributed in the early stage of healing after tendon injury, having the similar expression with mammals. Therefore, we hypothesized the bone marrow derived macrophage secreted miRNAs-containing exosomes play important functions in peritendinous adhesion after tendon injury. Overall design: we investigated miRNA expression profiles in exosomes from bone marrow derived macrophages by using high-throughput sequencing technology.

微小核糖核酸(MicroRNA)在复杂生理事件中,具备对不同生物学过程进行交叉调控与功能整合的潜力。本研究发现,骨髓源巨噬细胞外泌体(exosomes)中高表达的微小核糖核酸,可调控肌腱的纤维化愈合反应。值得注意的是,小鼠体内的纤维化相关微小核糖核酸-21(microRNA-21)在肌腱损伤后的愈合早期阶段呈现特异性表达分布,其表达模式与哺乳动物高度相似。据此,我们提出假说:骨髓源巨噬细胞分泌的携带微小核糖核酸的外泌体,在肌腱损伤后的肌腱周围粘连过程中发挥重要作用。整体实验设计:本研究采用高通量测序技术,对骨髓源巨噬细胞外泌体中的微小核糖核酸表达谱进行了检测分析。
