A mock community made of 17 bacterial species was assembled so that each species is represented by the same number of ribosomal operon sequences in the mix. This mock community was then used in a metabarcoding analysis using Nanopore long-read sequencing. Four primer sets targeting the ribosomal operon region were tested, and two different sequencing chemistries were used. The generated sequencing datasets were processed with the PRONAME pipeline, which allowed demonstrating how this pipeline can correct sequencing errors and significantly increase sequence accuracy.
本研究构建了由17种细菌组成的模拟群落(mock community),使混合体系中每种细菌的核糖体操纵子(ribosomal operon)序列数量保持一致。随后将该模拟群落应用于基于纳米孔(Nanopore)长读长测序的宏条形码测序(metabarcoding)分析。实验共测试了4组靶向核糖体操纵子区域的引物,并采用了两种不同的测序化学方案。所生成的测序数据集经PRONAME分析流程(PRONAME pipeline)处理后,证实该流程可有效校正测序错误并显著提升序列准确性。
