Study designed to identify genes that are induced in both BAT and skeletal muscle during acute adaptive thermogenesis in mouse using gene expression microarray. Animal studies were performed under approved UCLA animal research protocols and according to guidelines established in the Guide for Care and Use of Laboratory Animals. C57BL/6J mice were maintained in 12-h light/dark conditions and fed a regular chow diet (Purina 50010, Lab Diet, USA). Cold exposure was performed as described. After euthanasia, tissues were collected and frozen until use. Total RNA was isolated from mouse tissues by extraction with TRIzol from either brown adipose tissue (BAT) or skeletal muscle (quadriceps) after exposure to cold (4 degrees C for 4 hours). The mice were of the C57BL/6J strain and were maintained in 12-h light/dark conditions and fed a laboratory chow diet that consisted of 4.5% fat, 50% carbohydrate by weight (Lab Diet, Purina 50010).
本研究旨在通过基因表达微阵列技术,鉴定小鼠在急性适应性产热过程中,于棕色脂肪组织(BAT)与骨骼肌内均被诱导表达的基因。 本动物实验已获得加州大学洛杉矶分校(UCLA)动物研究伦理审批,并严格遵循《实验动物护理与使用指南》(Guide for Care and Use of Laboratory Animals)制定的相关规范开展。 实验所用C57BL/6J品系小鼠饲养于12小时光暗循环环境中,喂食常规维持饲料(美国Lab Diet公司,Purina 50010配方)。冷暴露实验按照已报道的方法实施。小鼠安乐死后采集组织并冻存备用。 冷暴露(4℃持续4小时)后,分别从棕色脂肪组织(BAT)与骨骼肌(股四头肌)中采用TRIzol试剂提取总RNA。 本实验所用小鼠为C57BL/6J品系,饲养于12小时光暗交替环境,所喂食的实验用维持饲料按重量百分比计含4.5%脂肪、50%碳水化合物(Lab Diet公司,Purina 50010配方)。
