Self-elongating neural tube organoids recapitulate key aspects of the morphology, anterior-posterior patterning, neural crest emergence and neural differentiation of mouse embryo in vivo by self-organization. We used single-cell RNA sequencing (scRNA-seq) to analyse the cell types and to reveal the sequence of transcriptional events in the emergence of neural crest cells and neural differentiation. Overall design: 10x single-cell RNA-seq of mouse neural tube organoids at day 6 (in duplicates), 7, 8, and 10.

自延伸神经管类器官可通过自组织过程,复现小鼠胚胎体内形态发生、前后轴模式建成、神经嵴发生及神经分化的关键特征。本研究采用单细胞RNA测序(single-cell RNA sequencing, scRNA-seq)技术分析细胞类群,并揭示神经嵴细胞发生与神经分化过程中转录事件的时序特征。实验整体设计:对培养第6、7、8、10天的小鼠神经管类器官开展10×单细胞RNA测序,其中第6天样本设置生物学重复。
