To investigate the BTK and GSK3 in the regulation of human B cell biology, we treated B cells from peripheral blood with either vehicle or BTK inhibitor or GSK3 in presence or absence of BCR cross-linking antibodies. We then performed gene expression profiling analysis using data obtained from RNA-seq of those samples at 24hrs. 为探究布鲁顿酪氨酸激酶(BTK)与糖原合成激酶3(GSK3)对人类B细胞生物学的调控作用,我们对外周血来源的B细胞开展如下处理:分别以溶媒对照、BTK抑制剂、GSK3抑制剂进行干预,并分别设置添加与不添加B细胞受体(BCR)交联抗体的实验组。随后,我们收集24小时后的样本,通过RNA测序(RNA-seq)获取数据,进行基因表达谱分析。
整体实验设计:对经BTK抑制剂或GSK3抑制剂处理的B细胞的RNA测序数据开展对比基因表达谱分析。
