Our study represents the first detailed analysis of thyroid hormone binding to the genome in the hindlimb of wild-type and TRa (-/-) tadpoles, with biological replicates, generated by ChIP-seq technology. The optimized data analysis workflows reported here should provide a framework for comparative investigations of TR binding profiles. Overall design: Examination of 3 technical replicate from the same RNA sample which extracted and merged from the several Xenopus tropicalis tadpoles.

本研究首次针对野生型与TRα(甲状腺激素受体α,Thyroid Hormone Receptor α)纯合敲除蝌蚪的后肢组织,开展甲状腺激素与基因组结合情况的详尽分析,实验设置生物学重复,相关数据由染色质免疫共沉淀测序(ChIP-seq, Chromatin Immunoprecipitation Sequencing)技术生成。本文报道的优化数据分析流程,可为甲状腺激素受体结合谱的比较研究提供通用框架。整体实验设计:针对同一份RNA样本开展3次技术重复,该RNA样本提取自多只热带爪蟾(Xenopus tropicalis)蝌蚪并经混合制备。
