To evaluate the effects of FEDB on NCCs induced in 2D conditions, we sampled 2D cultured cells and cell aggregates during FEDB treatment and compared gene expression patterns. Overall design: RNA sequencing profiles of 1231A3 iPSC-derived NCC aggregates, NCC aggregates treated with FEDB, CD271high+ NCCs sorted from 2D induction protocols (2D NCC CD271high), and 2D NCC CD271high treated with FEDB in 2D culture (2D 271H+FEDB).

为评估FEDB对二维(2D)培养条件下诱导的神经嵴细胞(Neural Crest Cells,NCCs)的调控作用,本研究于FEDB处理过程中采集二维培养细胞及细胞聚集体样本,并对其基因表达模式开展对比分析。整体实验设计:对以下四类样本进行RNA测序分析:1231A3诱导多能干细胞(induced pluripotent stem cell,iPSC)衍生的NCC聚集体、经FEDB处理的NCC聚集体、从二维诱导方案中分选获得的CD271高阳性NCCs(2D NCC CD271high),以及在二维培养体系中经FEDB处理的2D CD271high NCCs(2D 271H+FEDB)。
