Sialyl Transferase-EGFP transfected HeLa cells demonstrating the redistribution of the Golgi Apparatus to the Endoplasmic Reticulum upon treatment with Brefeldin A (BFA).
Images were collected as a 4D time series (z-stack over time). z-stacks of 48 individual timepoints are included in this collection of files as well as the movie SiEGFP_BFA.avi, which is the maximum projection of the z-stacks at the individual time points.
BFA was added according to the following protocol:
Timepoint1: Before addition of BFA 0seconds (8 Z-slices at 1 um apart)
Timepoint2: After addition of BFA 130seconds (8 Z-slices at 1 um apart)
Timepoint3: 238 seconds (11 Z-slices at 1 um apart)
Timepoint3 thru Timepoint48: Images captured every 5 seconds (11 Z-slices at 1 um apart at each time point)
Images were collected on a Carl Zeiss spinning disk confocal with a 63X 1.4 NA objective with 250 ms exposures at 5 s intervals.

本数据集采集得到四维时间序列(4D time series)图像,即随时间变化的z轴堆叠(z-stack)。本文件集中收录了48个独立时间点的z轴堆叠图像,同时附带视频文件SiEGFP_BFA.avi,该视频为各时间点z轴堆叠的最大投影(maximum projection)结果。
布雷菲德菌素A的添加遵循如下实验流程:
时间点1:布雷菲德菌素A添加前0秒(采集8层Z切片,层间距1 μm)
时间点2:布雷菲德菌素A添加后130秒(采集8层Z切片,层间距1 μm)
时间点3:处理后238秒(采集11层Z切片,层间距1 μm)
时间点3至时间点48:每5秒采集一次图像(每个时间点均采集11层Z切片,层间距1 μm)
图像采集使用卡尔蔡司(Carl Zeiss)转盘共聚焦显微镜(spinning disk confocal),搭配63× 1.4数值孔径(NA)物镜,曝光时长250 ms,采集间隔为5秒。
