The Janus liposozyme robustly eradicates infections and rapidly promotes wound closure and re-epithelialization on diabetic skin wound infected with methicillin-resistant S. aureus (MRSA). We used single cell RNA sequencing(scRNA-seq) to deep analyse local immune homeostasis manipulated by Janus on skin cells obtained from db/db mice . Overall design: db/db mice were infected with methicillin-resistant S. aureus (MRSA) and then were randomly divided into two groups: (i) PBS as control; (ii) TSeL with light irradiation (TSeL+L). Subsequently, skin tissues were harvested on day15 and further dissociated into single cells without separating dermis and epidermis. And skin cells were analyzed using scRNA-seq.
Janus脂质酶(Janus liposozyme)可强效根除感染,在耐甲氧西林金黄色葡萄球菌(methicillin-resistant S. aureus, MRSA)感染的糖尿病皮肤创面中快速促进创面闭合与再上皮化。我们采用单细胞RNA测序(single cell RNA sequencing, scRNA-seq),对从db/db小鼠获取的皮肤细胞中Janus所调控的局部免疫稳态开展深度分析。实验总体设计:将db/db小鼠先以耐甲氧西林金黄色葡萄球菌(MRSA)感染,随后随机分为两组:(i) 磷酸盐缓冲液(PBS)对照组;(ii) 光辐照TSeL组(TSeL+L)。后续于第15天收取皮肤组织,在未分离真皮与表皮的情况下将其解离为单细胞,随后通过scRNA-seq对皮肤细胞进行分析。
