Hematopoietic stem cells (HSCs) exhibit significant age-related phenotypic and functional alterations. Although single-cell technologies have elucidated age-related compositional changes, prospective identification of aging-associated HSC subsets has remained challenging. In this study, utilizing Clusterin (Clu)-GFP reporter mice, we demonstrated that Clu expression faithfully marks age-associated dysfunctional HSCs throughout life.

造血干细胞(Hematopoietic stem cells, HSCs)存在显著的年龄相关性表型与功能改变。尽管单细胞技术已阐明年龄相关的细胞组成变化,但前瞻性鉴定衰老相关的造血干细胞亚群仍颇具挑战。本研究利用簇集蛋白(Clusterin, Clu)-GFP报告基因小鼠,证实簇集蛋白的表达可在整个生命进程中精准标记衰老相关的功能异常造血干细胞。
