To investigate the function of CDCP1 in regulation of HCF proliferation, we cultured primary HCFs and transfected with non-targeting siRNA and CDCP1 siRNAs. Total RNA samples were extracted for RNA-seq to identify transcriptome-wide gene expression changes in the HCFs after CDCP1 knock-down (KD). Overall design: Comparative gene expression profiling analysis of RNA-seq data for HCF cells with and wihout CDCP1 KD.

为探究CDCP1在调控HCF增殖中的功能,我们培养了原代HCF,并分别转染非靶向小干扰RNA(small interfering RNA, siRNA)与CDCP1靶向siRNA。随后我们提取总RNA样本进行RNA测序(RNA-seq),以鉴定CDCP1敲低(Knock-down, KD)后HCF中全转录组水平的基因表达变化。实验整体设计:对存在CDCP1敲低与未发生CDCP1敲低的HCF细胞的RNA测序数据开展比较基因表达谱分析。
