To analyze the effect of type I IFN on global gene expression in differentiating monocytes, we employed microarray-based gene expression profiling. The MACS-purified mouse BM monocytes were cultured in M-CSF and treated with or without IFN for 60 h. The cells were harvested and isolated RNAs were subjected to microarray analyses. Overall design: MACS-purified monocytes from mouse bone marrow were divided into two groups (each in triplicates) and treated with 20 ng/mL M-CSF, together with or without 100 U of mouse IFN-beta for 60 h. The resulting cultures were harvested and the RNA samples were subjected to microarray analyses.

为探究I型干扰素(type I IFN)对分化中单核细胞的全局基因表达的影响,我们采用了基于微阵列的基因表达谱分析技术。经磁激活细胞分选(MACS)纯化的小鼠骨髓单核细胞,于巨噬细胞集落刺激因子(M-CSF)培养体系中培养,并分别施加干扰素处理与空白对照,处理时长为60小时。收集细胞后,提取的RNA样本将用于微阵列分析。 总体实验设计:将经MACS纯化的小鼠骨髓单核细胞分为两组(每组设置3次生物学重复),两组均添加20 ng/mL的M-CSF,其中一组额外加入100 U小鼠β干扰素(IFN-β),共同培养60小时。收集两组培养物,提取RNA样本后进行微阵列分析。
