By using a mixed-ligand approach, two new coordination polymers with the chemical formula of {[Co<sub>2.5</sub>(nbta)(trz)<sub>2</sub>(H<sub>2</sub>O)]·3H<sub>2</sub>O}<sub>n</sub> (<b>1</b>, H<sub>3</sub>nbta = 5-nitros,2,3-benzenetricarboxylic acid and Htrz = 1<i>H</i>-1,2,4-triazole) and [Cu<sub>3</sub>(tib)<sub>2</sub>(TATB)<sub>2</sub>]·DMF·2H<sub>2</sub>O (<b>2</b>, tib = 1,3,5-tris(1-imidazolyl)benzene and H<sub>3</sub>TATB = 4,4′,4″-s-triazine-2,4,6-triyltribenzoic acid) were constructed using a solvothermal method by reaction of metal salts with the corresponding carboxylate and the N-donor co-ligands. After that, they were used for the treatment of acute lung injury animal model. The content of the IL-18 and TNF-α released by the alveolar macrophages was evaluated with enzyme-linked immunosorbent assay (ELISA) detection assay. The activation level of the NF-κB was measured with real-time RT-PCR.
采用混合配体策略,通过溶剂热法使金属盐与相应的羧酸配体及氮给体共配体发生反应,合成了两例新型配位聚合物,其化学式分别为{[Co₂.₅(nbta)(trz)₂(H₂O)]·3H₂O}<sub>n</sub>(化合物<b>1</b>,其中H₃nbta=5-硝基-1,2,3-苯三甲酸,Htrz=1<i>H</i>-1,2,4-三唑)以及[Cu₃(tib)₂(TATB)₂]·DMF·2H₂O(化合物<b>2</b>,其中tib=1,3,5-三(1-咪唑基)苯,H₃TATB=4,4′,4″-均三嗪-2,4,6-三基三苯甲酸)。随后将该两例配位聚合物应用于急性肺损伤动物模型的干预治疗。采用酶联免疫吸附测定(enzyme-linked immunosorbent assay, ELISA)法检测肺泡巨噬细胞释放的白细胞介素18(IL-18)与肿瘤坏死因子α(TNF-α)的含量;通过实时逆转录聚合酶链反应(real-time RT-PCR)测定核因子κB(NF-κB)的激活水平。
