KRAS突变型直肠腺癌肿瘤组织中,基质与细胞外基质(extracellular matrix)相关基因呈现表达下调趋势。本研究采用Affymetrix U133芯片平台,对KRAS野生型与KRAS突变型直肠癌标本的转录组(transcriptome)差异进行解析。研究样本取自通过TIMING试验(ClinicalTrials.gov,编号NCT00335816)招募的局部晚期直肠癌患者,共收集76份微卫星稳定(microsatellite stable)的治疗前活检样本并提取总RNA,随后基于KRAS突变状态完成差异基因表达分析。
