Single cell RNA sequencing was performed on CD45+Ly6G-CD64+MerTK+ macrophages isolated from the lungs of mice 5 weeks after transplantation of 4T1-Luc mammary gland tumors. The purpose of this study was to determine the unique macrophage subsets present in the lungs of mice bearing mammary tumor metastases. Overall design: Mice received injections of 10k 4T1-Luc cells into the #4 mammary gland. Tumors were allowed to progress for 5 weeks, at which point the lungs were harvested. The lungs were digested and macrophages were sorted by FACS, followed by single cell RNA sequencing.
本研究针对移植4T1-Luc乳腺肿瘤5周后的小鼠肺部分离得到的CD45+Ly6G-CD64+MerTK+巨噬细胞,开展了单细胞RNA测序(single cell RNA sequencing)。本研究旨在明确携带乳腺肿瘤转移灶的小鼠肺部中存在的独特巨噬细胞亚群。整体实验设计:向小鼠第4乳腺脂肪垫注射10⁴个4T1-Luc细胞,待肿瘤生长5周后收取肺部组织;对肺部组织进行酶消化后,通过荧光激活细胞分选术(Fluorescence-Activated Cell Sorting, FACS)分选巨噬细胞,随后进行单细胞RNA测序。
