该数据集为2021年至2024年,在青藏高原玉珠峰、扎当冰川等16条冰川,纳木错、普莫雍错等30个湖泊,以及林芝、保山等6个城市乡镇等采集的冰川冰雪、冰芯、、冰前土壤、湖水、河流等生境采集的样品中分离得到的纯菌菌株信息及相应的纯菌菌落图片。附有一个表格信息,包括该菌株的种属名、基因序列号、保藏时间、分离该菌株的样品来源地和生境信息,以及分离该菌株所用的培养基类型、培养温度等。表格中”-”表示没有该信息。这些纯菌菌落呈现不同的颜色,这可能与微生物适应青藏高原强辐射、低温、寡营养的环境有关,通过产生不同的色素等来适应这一严酷的环境。这些纯菌是通过稀释平板涂布,在不同温度下培养(培养基为R2A等)分离,确定无其他杂菌污染、为单菌落之后获得。得到的单菌落后续会进行16S rRNA全长测序,通过97%的相似度确定其是否为新的种;如果该序列与最相似菌株的序列相似度低于97%,会进行全基因组测区,确定该菌株在相应的属、科甚至门类中的系统发育关系,确定该菌株是否具有新的系统发生地位。这些纯菌的分离和获得进一步开发了青藏高原的微生物资源库,为充分利用这些宝贵的微生物资源提供了理论基础。

This dataset contains pure bacterial strain information and corresponding pure bacterial colony images isolated from samples collected from habitats such as glacial snow and ice, ice cores, proglacial soil, lake water, and rivers in 16 glaciers including Yuzhu Peak and Zhadang Glacier, 30 lakes including Nam Co and Pumoyong Co, and 6 cities and towns including Nyingchi and Baoshan on the Qinghai-Tibet Plateau from 2021 to 2024. It is accompanied by a table containing information such as the species and genus name of the strain, gene sequence number, preservation time, sample origin and habitat information from which the strain was isolated, as well as the type of medium used for isolation and cultivation temperature. A "-" in the table indicates no such information. These pure bacterial colonies exhibit different colors, which may be related to the adaptation of microorganisms to the environment of strong radiation, low temperature, and oligotrophic conditions on the Qinghai-Tibet Plateau, where they adapt to this harsh environment by producing different pigments. These pure bacteria were isolated via dilution plating and cultured at different temperatures (using media such as R2A), and obtained after confirming no contamination by other mixed bacteria and being single colonies. The obtained single colonies were subsequently subjected to full-length 16S rRNA sequencing to determine whether they are new species based on a 97% similarity threshold. If the sequence similarity with the most similar strain is less than 97%, whole-genome sequencing will be performed to determine the phylogenetic relationship of the strain at the genus, family, or even phylum level, and to confirm whether it has a new phylogenetic status. The isolation and acquisition of these pure bacteria have further developed the microbial resource bank of the Qinghai-Tibet Plateau and provided a theoretical basis for the full utilization of these valuable microbial resources.
