This study analyzed airborne bacteria communities in expansive underground buildings via high-throughput sequencing of 16srDNA V4 fragment. In this study, we employ a meticulous approach to collect biological microbe samples using cotton swabs that have been pre-immersed in the 1-ml Liquid Amies preservation medium of Eswabs to ensure the maximum capture of biomass. The sampling process, conducted from September 2023 to August 2024 across 44 distinct sites and a total of 452 DNA samples and 36 RNA samples within the science buildings at XJTLU, includes a variety of environmental surfaces, such as vents.

本研究针对大型地下建筑中的空气传播细菌群落展开分析，采用16S rDNA V4区片段（16srDNA V4 fragment）高通量测序技术完成检测。本研究采用严谨规范的采样策略：使用预先浸润于1 mL Amies液体保存培养基的Eswabs拭子采集微生物样本，以最大化捕获生物量。本次采样于2023年9月至2024年8月间在西交利物浦大学（XJTLU）理科楼内的44个不同点位开展，共采集452份DNA样本与36份RNA样本，采样覆盖通风口等多种环境表面。
