p85beta of the PI3K can localize to the nucleus. Our data showed that it can increase the expression of a transcription factor BCLAF1. To determine whether p85beta has any affect on gene transcription and to identify gene targets that are regulated through BCLAF1, RNAseq was performed on p85beta-expressing cells transfected with BCLAF1 siRNA . Transient transfection of BCLAF1 siRNA or mock transfection for 72 hours in DOV13 cells stably expressing p85beta. Stable expression of empty vector was control. Three biological replicates per condition.
磷脂酰肌醇3-激酶(PI3K)的p85β亚基可定位于细胞核内。本研究数据显示,该亚基可上调转录因子BCLAF1(BCL2-associated transcription factor 1)的表达水平。为明确p85β亚基是否对基因转录存在调控作用,并筛选受BCLAF1调控的基因靶点,研究人员对过表达p85β亚基且转染了BCLAF1小干扰RNA(siRNA)的细胞开展了RNA测序(RNAseq)。实验采用稳定过表达p85β亚基的DOV13细胞,分别进行BCLAF1 siRNA瞬时转染与空白mock转染,处理时长为72小时;以仅转染空载体的稳定细胞株作为对照组。每组实验设置3次生物学重复。
