In order to find function of TTHA1634 protein from T. thermophilus HB8, we observed the expression profile of the total mRNA of the TTHA1634-deficient strain grown in a rich medium at 70°C for 420, and 600 min. Two TTHA1634-deficient T. thermophilus HB8 strains were each pre-cultured at 70°C for 16 h in 3 mL of TT medium containing 0.8% polypeptone, 0.4% yeast extract, 0.2% NaCl, 0.4 mM CaCl2, and 0.4 mM MgCl2, which was adjusted to pH 7.2 with NaOH. The cells (2 mL) were inoculated into 1 L of the same medium and then cultivated at 70°C. Cells were collected after 420, and 600 min, respectively, and then, the expression of each mRNA was analyzed.

为探究嗜热栖热菌HB8(Thermus thermophilus HB8)中TTHA1634蛋白的功能,我们对在丰富培养基中于70℃下分别培养420分钟、600分钟的TTHA1634缺陷菌株的总信使RNA(mRNA)表达谱进行了观测。将两株TTHA1634缺陷型嗜热栖热菌HB8菌株分别接种于3 mL含0.8%蛋白胨、0.4%酵母提取物、0.2%氯化钠、0.4 mM氯化钙及0.4 mM氯化镁的TT培养基中,经NaOH调节pH至7.2后,于70℃预培养16小时。取2 mL菌液接种至1 L相同培养基中,随后于70℃继续培养。分别在培养420分钟、600分钟后收集菌体,进而对各mRNA的表达情况进行分析。
