Microcosm experiments were performed to investigate the effect of organic material composition on soil bacterial and archaeal communities under paddy soil conditions. Rice straw (RS) and farmyard manure (FM) were used as the original organic materials, and three types of residues were prepared by sequential extraction from the original materials (R1RS and R1FM, lipid extracted organic materials; R2RS and R2FM lipid and sugar extracted organic materials; R3RS and R3FM, lipid, sugar, and hemicellulose extracted materials). The organic materials were applied to soil microcosms at a 5 or 10 Mg/ha level. Soil microcosms were incubated under flooded conditions, and DNA was extracted at 0, 1, 2, 4, 8, and 16 weeks after the incubation.
本研究开展微宇宙实验(microcosm experiment),以探究稻田土壤条件下有机质组分对土壤细菌与古菌群落的影响。本研究选取水稻秸秆(RS)与农家肥(FM)作为原始有机质材料,通过对原始物料进行连续萃取制备得到三类残余组分:R1RS与R1FM为经脂质萃取所得的有机质;R2RS与R2FM为经脂质与糖类萃取所得的有机质;R3RS与R3FM为经脂质、糖类与半纤维素萃取所得的有机质。将上述有机质以5 Mg/ha或10 Mg/ha的施用量施加至土壤微宇宙体系中。土壤微宇宙体系于淹水条件下培养,并在培养后的0、1、2、4、8及16周时提取基因组DNA。
