To date, there have been limited high quality libraries of cardiomyocyte maturation during the perinatal period, in part owing to the difficulty of isolating large perinatal cardiomyocytes. We previously developed a method utilizing large-particle fluorescent activated cell sorting (LP-FACS) to isolate adult cardiomyocytes for single cell RNA-seq (Kannan et al., Circ Res, 2019). We utilize this method to generate a reference of perinatal cardiomyocyte maturation. We isolated CMs from mice at the following timepoints using LP-FACS: e14, e18, p0, p4, p8, p11, p14, p18, p22, p28, p35, p56.

迄今为止,围产期心肌细胞(cardiomyocytes)成熟相关的高质量转录组文库仍较为稀缺,这在一定程度上源于难以分离获取大型围产期心肌细胞。我们此前曾开发一种利用大颗粒荧光激活细胞分选(large-particle fluorescent activated cell sorting, LP-FACS)技术的方法,用于分离成年心肌细胞以开展单细胞RNA测序(single cell RNA-seq)研究(Kannan 等,《循环研究》(Circ Res),2019 年)。本研究沿用该方法,构建围产期心肌细胞成熟的参考数据集。我们通过LP-FACS技术从小鼠体内分离得到心肌细胞(cardiomyocytes, CMs),采集的时间点包括:e14、e18、p0、p4、p8、p11、p14、p18、p22、p28、p35及p56。
