We observed an increasing trend of nuclear SLC29A2 expression associated with high histological de-differentiation grades, metastasis and poor patient prognosis by immunohistochemical analysis of HCC tumors. Treatments of dipyridamole to SLC29A2 expressing SK-hep1 cells reduced cell viability and cell migration. To examine the anticancer activities of dipyridamole, transcriptome analysis was performed to detect the modulations of oncogenic SLC29A2 and the downstream signaling pathways. Gene expression profiles of dipyridamole and/or sorafenib treatment in SLC29A2-expressing Sk-hep1 cells were generated by Illumina RNA sequencing and compared to profiles derived from mock control cells.

本研究通过对肝细胞癌(Hepatocellular Carcinoma, HCC)肿瘤组织开展免疫组化分析,发现细胞核内SLC29A2的表达水平随组织学去分化分级升高、肿瘤转移及患者不良预后呈现上升趋势。将双嘧达莫(dipyridamole)作用于表达SLC29A2的SK-hep1细胞后,可降低细胞存活率与迁移能力。为探究双嘧达莫的抗癌活性,本研究通过转录组分析检测了致癌性SLC29A2及其下游信号通路的调控变化。本研究采用Illumina RNA测序技术,获取了经双嘧达莫、索拉非尼(sorafenib)单独或联合处理的表达SLC29A2的SK-hep1细胞的基因表达谱,并与空白对照细胞的基因表达谱进行了对比分析。
