We analysed genome wide DNA methylation of mature (CD34-CD15+) and immature (CD34+CD15-) hematopoietic cells from patients with chronic phase CML at diagnosis. before any treatment. and compared it to their counterpart cells isolated from healthy donors. Overall design: Bisulfite converted DNA of mature (CD34-CD15+) and immature (CD34+CD15-) cells from healthy donors and CML patients was hybridized to the Infinium Human Methylation 450K BeadChip. DNA bisulfite conversion and hybridization on the arrays were performed by Integragen. SA (Evry. France) following the Illumina Infinium HD methylation protocol.
本研究对确诊时未接受任何治疗的慢性期慢性髓系白血病(chronic phase CML)患者的成熟(CD34-CD15+)与未成熟(CD34+CD15-)造血细胞开展了全基因组DNA甲基化(genome wide DNA methylation)水平分析,并将其与健康供体分离的对应细胞进行了比对。
整体实验设计:将健康供体及慢性期CML患者的成熟(CD34-CD15+)与未成熟(CD34+CD15-)细胞的亚硫酸氢盐转化DNA(bisulfite converted DNA),采用Infinium Human Methylation 450K BeadChip(人类甲基化450K微珠芯片)进行杂交检测。DNA亚硫酸氢盐转化及芯片杂交实验由法国埃夫里市的Integragen公司按照Illumina Infinium HD甲基化实验方案完成。
