Study designed to identify genes that are induced in both BAT and skeletal muscle during acute adaptive thermogenesis in mouse using gene expression microarray. Animal studies were performed under approved UCLA animal research protocols and according to guidelines established in the Guide for Care and Use of Laboratory Animals. C57BL/6J mice were maintained in 12-h light/dark conditions and fed a regular chow diet (Purina 50010, Lab Diet, USA). Cold exposure was performed as described. After euthanasia, tissues were collected and frozen until use. Total RNA was isolated from mouse tissues by extraction with TRIzol from either brown adipose tissue (BAT) or skeletal muscle (quadriceps) after exposure to cold (4 degrees C for 4 hours). The mice were of the C57BL/6J strain and were maintained in 12-h light/dark conditions and fed a laboratory chow diet that consisted of 4.5% fat, 50% carbohydrate by weight (Lab Diet, Purina 50010).
本研究旨在通过基因表达微阵列(gene expression microarray)技术,鉴定小鼠在急性适应性产热过程中,棕色脂肪组织(BAT)与骨骼肌内均被诱导表达的基因。本动物实验已按照加州大学洛杉矶分校(UCLA)批准的动物研究规程开展,并严格遵循《实验动物护理与使用指南》(Guide for Care and Use of Laboratory Animals)规定的操作规范。实验采用C57BL/6J品系小鼠,饲养于12小时光暗循环环境中,饲喂由4.5%脂肪、50%碳水化合物(按重量占比)组成的常规实验饲料(Lab Diet, Purina 50010,美国产)。冷暴露处理参照既定方案实施,将小鼠置于4℃环境中持续4小时。待小鼠实施安乐死后采集组织样本,冷冻保存直至后续实验使用。采用TRIzol试剂分别从冷暴露后的棕色脂肪组织及骨骼肌(股四头肌)中提取总RNA。
