In this experiment the relationship between SOX2 levels and chromatin accessibility was assayed using ATAC-seq during embryonic stem cell differentiation. A doxycycline inducible cell line (SOX2 TetON) was employed to experimentally control SOX2 expression. Overall design: Transposase accessible chromatin profiles were generated at high read depth in three experimentall conditions. Sox2 TetON ES cells differentiated in the presence or absence of 1ug/ml doxycycline were compared to differentiated WT epiblast progenitor cells.
本实验在胚胎干细胞分化过程中,采用转座酶可及性测序(ATAC-seq)检测SOX2表达水平与染色质可及性之间的关联。本研究使用多西环素诱导型细胞系(SOX2 TetON),通过实验手段调控SOX2的表达水平。整体实验设计:在三种实验条件下以高测序深度生成转座酶可及染色质图谱。将在1微克/毫升多西环素存在与不存在条件下分化的SOX2 TetON胚胎干细胞,与已分化的野生型(Wild Type)上胚层祖细胞进行对照比较。
